چکیده

Recent work from this laboratory has been concerned with elucidating the function of the non-thrombin-forming half or propiece of prothrombin in the zymogen activation process. These studies have demonstrated that Fragment 1, the NH2-terminal region of the propiece (Fragment 1.2), is responsible for the Ca2+-mediated binding of prothrombin to phospholipid vesicles (1) and that Fragment 2, the COOH-terminal region of the propiece, is required for acceleration of prothrombin activation by Factor V,i (2). Investigations of the kinetic consequences of the functioning of the two propiece regions (3) have led to the following proposal for the principal pathway of prothrombin conversion to thrombin.’

Frequency of Factor V Leiden and Prothrombin Polymorphism in South of Iran

Normal hemostasis requires balanced regulation of prothrombotic and antithrombotic factors. Inherited alteration of factor V and prothrombin gene, the G20210A mutation, increases the resistance of factor V to degradation and booster production of prothrombin respectively. These alterations can increase hypercoagulability leading to thrombotic consequences. We aimed to assess the frequencies of ...

Prothrombin is activated on vascular endothelial cells by factor Xa and calcium.

Vascular endothelial cells derived from adult bovine aorta (ABAE) treated with factor Xa and calcium were found to activate prothrombin. In contrast, nonvascular cells (human foreskin fibroblasts, bovine corneal endothelial cells, or human fetal lung cells) had either no or very little effect on prothrombin activation. In the presence of 6 X 10(5) ABAE cells, 20 ng of factor Xa converted 90 mic...

Frequency of factor V Leiden (G1691A) and prothrombin (G20210A) polymorphisms in Population of Kerman Province, Iran

Background & Aims:Thromboembolism is an acute cardiovascular disease that ranges from clinically unimportant to massive embolism. Both acquired and hereditary risk factors contribute to the disease.
We aimed to determine the prevalence of two hereditary predisposing factor of the disease, prothrombin G20210A and factor V Leiden (G1691A) polymorphisms, in Kerman population.<br /...

Factor V Leiden, MTHFR C677T and Prothrombin Gene Mutation G20210A in Iranian Patients with Venous Thrombosis

Background: Factor V Leiden, Prothrombin gene (G20210A) and MTHFR (C677T) polymorphism are the main biomarkers for evaluation of tendency for venous thromboembolism. We aimed to investigate the frequency of mutations in factor V Leiden, Prothrombin G20210A and MTHFR C677T and identify the genetic status for these mutations in patients with venous thrombosis. Methods: This study was carried out...

The Conversion of Prothrombin to Thrombin

Prothrombin is activated by activated Factor X f (Factor V and phospholipid) in the presence of diisopropylfluorophosphate which preferentially inhibits thrombin. Under these conditions a single activation fragment, designated Fragment 1.2, is released which represents the complete nonthrombin portion of the prothrombin molecule. The same fragment is seen as a transient species in the absence o...
